Rovers started well with Gordon Murphy setting up Jonny Wilson in the first minute but the winger failed to take the chance.
Ellon showed that they could be dangerous in the 11th minute when a cross from the right found their centre forward for a free header but Carlukes new keeper Jonny Grier was on hand with a good save from thee close range header.
This save proved to be crucial as in the next minute Rovers went on the attack with Murphy setting Ian Watt free, Watt waltzed round three defenders before hitting a low drive in to the corner of the net for 1-0 to the visitors from Carluke.
Carluke continued to dominate the match and doubled their lead in the 24th min when good play through the midfield set Wilson free on the right, his cut back to Watt saw the striker hit a shot on goal that fell to the feet of strike partner Greig Cavanagh who controlled well before rifling a shot in to the Ellon net for 2-0 to the Rovers.
Cavanagh also had the next chance for Carluke when he was set up by a free kick from Watt but his effort was turned away for a corner that came to nothing.
Ellon had a rare chance on the half hour mark when their centre forward turned away from the Rovers defence after a throw in but his rasping shot went across Griers goal and past the far post.
Rovers dominated what was left of the first half from this point but chances for Watt and Wilson came to nothing and they went in at half time content with a 2-0 lead.
The second half saw Carluke come out of the traps flying and increased their lead further in the 48th minute when a cross from the right by Dean Kindlan found the waiting Murphy who headed home for 3-0.
Carluke were well and truly in control of this game now and put it out of sight of their hosts after 72 mins when Murphy played a long ball to Cavanagh to chase down and he did this well beating a home defender before slotting the ball home from a tight angle for 4-0.
Watt showed great control a minute after this goal taking down a high ball but his parting shot did not match his control and the chance was lost.
The home side had a late flurry and they got a goal back in the 85th min when a free kick played to the back post was headed across goal before ending in the Rovers net for 4-1.
The last couple of minutes saw the home side have more efforts on goal than they had in the rest of the match but Grier in the Rovers goal brought of a couple of fine saves to keep the score at 4-1 to Carluke and send them in to the next round.
